ABSTRACT

Objective: To observe the clinical effects of bevacizumab in the treatment of familial epistaxis caused by hereditary hemorrhagic telangiectasia (HHT). Methods: The data of 27 patients with familial epistaxis caused by HHT who were treated with bevacizumab intravenously from Beijing Anzhen Hospital, the First Clinical Center of Chinese People's Liberation Army General Hospital and Binzhou Central Hospital between December 2016 and December 2019 were retrospectively analyzed. There were 14 males and 13 females, aged (55.3±11.2) years. The dose of bevacizumab was calculated according to the body weight of 5 mg/kg. The curative effect was observed one month after the first treatment. Visual analogue scale (VAS) was used to compare patients' self-scores of systemic symptoms before and after treatment. Epistaxis severity score (ESS) was used to compare and analyze the six problems (including the frequency, duration, intensity, treatment demand, anemia and blood transfusion) of the patients before and after treatment. The changes of hemoglobin levels before and after treatment were compared. SPSS 20.0 statistical software was used to process the data. Results: Among the 27 patients at one month after the first bevacizumab treatment, 22 cases reported that the severity of epistaxis was improved significantly, and 5 cases reported that the treatment effect was not significant. The effective rate was 81.5% (22/27). The significant effect in 22 patients lasted for 5-24 months, with a median duration of 11.23 months. The VAS score of systemic symptoms decreased significantly compared with that before treatment (2.41±2.55 vs 8.19±1.47, t=9.708, P<0.01). The scores of six aspects and standardized scores of ESS were significantly decreased after treatment (epistaxis frequency: 1.78±1.22 vs 3.44±0.80, t=6.814, P<0.01; epistaxis duration: 0.85±0.91 vs 3.00±0.73, t=8.845, P<0.01; epistaxis intensity: 0.19±0.40 vs 1.00±0.00, t=10.696, P<0.01; treatment demand: 0.22 ± 0.42 vs 1.00±0.00, t=9.539, P<0.01; anemia: 0.41±0.50 vs 0.89±0.32, t=4.914, P<0.01; blood transfusion: 0.11±0.32 vs 0.41±0.50, t=3.309, P<0.01; ESS standardized score: 2.50±2.45 vs 7.60±1.30, t=9.344, P<0.01). The hemoglobin level after treatment was significantly higher than that before treatment ((105.48±24.31) g/L vs (73.07±23.71) g/L, t=6.864, P<0.01). Among the 27 patients, there were 8 cases of HHT1 (ENG gene) and 19 cases of HHT2 (ACVRL1 gene). The improvement duration of epistaxis in group HHT1 and group HHT2 was (4.76±5.12) months and (7.60±10.84) months, respectively, which was in group HHT2 longer than that of group HHT1, but there was no significant difference between the two groups (P>0.05). There was no significant difference in ESS scores between the two groups before and after treatment (P>0.05). Two female patients had amenorrhea after the first medication. All patients had no other adverse reactions and complications. Conclusion: Intravenous bevacizumab is significantly effective and safe in the treatment of familial epistaxis caused by HHT.

ABSTRACT

OBJECTIVE@#To observe the effect of pretreatment of acupuncture on the expression of nucleotide-binding oligomerization domain-like receptor 3(NLRP3), Caspase-1, interleukin1β(IL-1β) and the number of activated microglia (MG) in the hippocampus in Alzheimer's disease (AD) like rats, so as to explore the mechanism of pretreatment of acupuncture in preventing and treating AD.@*METHODS@#A total of 36 SD rats were randomly divided into a blank group, a model group and an electroacupuncture (EA) group, 12 rats in each group. The AD like rat model was established by 8-week continuous intraperitoneal injection of D-galactose (120 mg·kg@*RESULTS@#Compared with the blank group, the average escape latency was prolonged (@*CONCLUSION@#Pretreatment of acupuncture could prevent and treat the learning-memory dysfunction in AD like rats, and its mechanism may be related to the inhibition of NLRP3 inflammatsome related protein and MG activation.

Autophagy is a highly conserved physiological and biochemical process in which organisms maintain homeostasis of the cellular environment. Autophagy is widely involved in the occurrence and development of many diseases, especially in inflammatory diseases, which is a hot topic in recent years. Autophagy-related genes (ATGs) play important roles in regulating autophagy in many aspects, thus affecting the process and prognosis of inflammatory diseases. This article reviews the regulatory roles and mechanisms of autophagy in inflammatory diseases, as well as the functions and roles of several important ATGs in inflammatory diseases.

ABSTRACT

OBJECTIVE@#To observe the therapeutic effect of pre-acupuncture at Neiguan (PC 6) and Zusanli (ST 36) on exercise-induced fatigue.@*METHODS@#A total of 50 subjects were divided into an observation group and a control group by random number table method, 25 cases in each one. In the observation group, acupuncture was applied at Neiguan (PC 6) and Zusanli (ST 36) before exercise, once a day, consecutive 5-day treatment was required. In the control group, there was no intervention. The subjective fatigue perception score, serum levels of lactate and lactate dehydrogenase, heart rate and oxygen saturation before and after exercise were observed in the two groups.@*RESULTS@#Compared before exercise, the subjective fatigue perception scores, serum levels of lactate and lactate dehydrogenase and heart rates after exercise were increased in the two groups (0.05).@*CONCLUSION@#Pre-acupuncture at Neiguan (PC 6) and Zusanli (ST 36) can effectively reduce the subjective fatigue perception score,serum levels of lactate and lactate dehydrogenase and heart rate to improve exercise-induced fatigue.

ABSTRACT

Objective To study the X-ray signs of forearm and leg in skeletal fluorosis and its diagnostic value,aim at finding the easy examination parts.Methods One thousand four hundred and forty subjects were examined using developed shield,darkroom and other portable dedicated device combined with a small X-ray machine.A total of 384 cases were diagnosed skeletal fluorosis.All patients were divided into different groups and the time,degree and range of X-ray to the forearm and calf elbow,knee,and long bone were compared.Results The X-ray change in the forearm elbow was earlier than that of the leg knee,and trabecular bone change was the earliest indicator,197 cases and 157 cases,respectively,and the difference was statistically significant (x2 =28.006,P < 0.01).Membrane ossification of forearm backbone was earlier than that of the leg,and most of them were degree Ⅰ photos,213 cases and 126 cases respectively.The difference was statistically significant (x2 =17.626,P < 0.01).The direction of the interosseous membrane ossification was from the forearm radius to the ulna,then to the fibula and tibia,and was accompanied by changes in the aggravation of forearm.A variety of indicators were observed,especially the membrane ossification in bone and joint trabecular bone and the long bone was the most active,and the forearm was more sensitive,obviously than that of the calf.Conclusion In the X-ray screening or detection of endemic fluorosis,the forearm radiography is a simple,economical,and effective diagnostic method.

ABSTRACT

Objective To explore the application value of single intensifying screen cassette in examination of Kashin-Beck disease through X-ray photographing.Methods Single intensifying screen cassette and traditional changing bag with exposure conditions were used to check right wrists of 110 children in Kashin-Beck disease areas.The FJ personal dosimeter was used to measure exposure dose.Photo graphing quality and diagnosis effect were assessed.Results The radiation dose of children in single intensifying screen cassette group was (207 ± 39)μsv/h,while in the traditional changing bag group was (1425 ± 63)μsv/h.The difference between the two groups was statistically significant(t =140.16,P < 0.05).The high-quality photograph rate of the two methods was 94.55% (104/110) and 92.73% (102/110),respectively,and the difference was not statistically significant(P > 0.05).The good photo rate in the single intensifying screen cassette was 96.36% (94/110),which was significantly higher than that of the traditional changing bag group[44.55% (49/110),x2 =70.92,P < 0.01].Conclusions X-ray radiation dose in single intensifying screen cassette group is smaller than that of the traditional changing bag group,and the image quality of the radiograph of the new method is also superior.It has a good practical value in the X-ray examination of Kashin-Beck disease.

ABSTRACT

<p><b>OBJECTIVE</b>To research the bacteriostatic effects of Qingkailing Injection Extract (QKLIE) and combination therapy of Qingkailing Injection (QKLI) and antibiotics on bacteria carrying New Delhi metallo-3-lactamase 1 (NDM-1) blaNDM-1 resistance gene, and to determine their minimal inhibitory concentrations (MIC).</p><p><b>METHODS</b>The antimicrobial experiments of QKLIE (Radix Isatidis, baicalin, gardenia, honeysuckle) and combination therapy of QKLI and antibiotics were performed by using the agar dilution method and K-B method. The MIC was determined from each extract.</p><p><b>RESULTS</b>There were different degrees of inhibitory effects on resistant bacteria carrying blaNDM-1 by extracts from main components of QKLI. Of them, the inhibitory effect of baicalin was the best and the MIC of the resistant bacteria was 0.015 g/mL to WD, 0.020 g/mL to WX, 0. 005 g/mL to WJ, and more than 0.020 g/mL to pGEX-4T-NDM-1/DH5alpha (GST-NDM-1), respectively. The MIC value of each extract was sequenced from high to low as baicalin, honeysuckle, gardenia, and Radix Isatidis. Furthermore, combination therapy of QKLI and antibiotics greatly enhanced the antimicrobial activity of each antibiotics when used alone, showing very obvious antibacterial effects on multidrug resistant bacteria carrying blaNDM-1 gene. Of them, the optimal effects were obtained when combined with penicillins (penicillin G, mezlocillin, piperacillin/ tazobactam, ampicillin/sulbactam), with the antibacterial effects improved by 10 folds. The antibacterial effects of other kinds of antibiotics were improved to some extent. Conclusions QKLIE and combination therapy of QKLI and antibiotics showed better bacteriostatic effects on resistant bacteria carrying blaNDM-1 gene. This study provided theoretical bases for drug development, medication and treatment for super-resistant bacteria carrying blaNDM-1.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To assess the validity of the lateral gutter drive-through (LGDT) test in diagnosing posterolateral rotational instability (PLRI) of the knee joint.</p><p><b>METHODS</b>Between October 2009 and February 2012, 115 consecutive patients were enrolled into this prospective diagnostic study. The tibia external rotation dial test was used as the gold standard for diagnostic reference. According to the dial test, the patients were divided into a study group (35 patients) and a control group (80 patients). The LGDT test was performed on all patients during arthroscopic surgery. The sensitivity and specificity of the LGDT test were calculated. The statistical difference of sensitivities in patient subgroups defined by injury patterns was tested using χ(2) test. And the correlation between the extent of tibia external rotational instability and the sensitivity of LGDT test was analyzed using logistic regression.</p><p><b>RESULTS</b>The sensitivity and specificity of the LGDT test were calculated as 91.4% and 93.8%, respectively. The sensitivity of detection for acute PLRI was 9/10 vs. 92.0% in chronic cases (χ(2) = 0.036, P = 0.849). Popliteus femoral "peel off" lesions were detected with a sensitivity of 100% vs. 87.0% in cases of non "peel off" lesions (χ(2) = 1.712, P = 0.536). The sensitivity of detecting isolated external rotational instability vs. combined instability (rotational and varus) was 90.5% and 13/14, respectively (χ(2) = 0.062, P = 0.805). The sensitivity of the LGDT test was correlated with the extent of tibial external rotational instability (r = 1.000, P = 0.011).</p><p><b>CONCLUSIONS</b>The LGDT test is a reliable method to diagnose PLRI of the knee joint. The highest sensitivity is observed for patients with the femoral "peel off" injury pattern. The sensitivity of the LGDT test is correlated with the extent of tibia external rotational instability.</p>

ABSTRACT

A patient with both a femoral attachment injury (peel-off injury) and musculotendinous junction avulsion of the popliteus, a so-called “floating popliteus tendon injury”, received arthroscopy-assisted popliteus reconstruction. The injured ligaments were addressed in the same procedure, including mini-open direct repair of the femoral avulsed fibular collateral ligament (FCL), suture repair of the grade 3 medial collateral ligament (MCL), and reconstruction of the posterior cruciate ligament (PCL).

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the clinical significance of arthroscopic anterior cruciate ligament (ACL) reconstruction using the remnant-preserved technique.</p><p><b>METHODS</b>From August 2008 to September 2009, 70 cases with the remnant of injured ACL were included in the trials, which were randomized into the remnant preservation (RP) group and the control group, 35 cases in each group. All patients in the two groups underwent arthroscopic ACL reconstruction surgeries, with ACL-remnant preserving technique in RP group and ACL-remnant resection in control group, respectively. The injured ACL was reconstructed with allograft in all cases. Postoperative follow-up assessment included the International Knee Documentation Committee (IKDC) grading and Lysholm score, Lachman test, pivot shift test and KT-1000 measurement, proprioception measurements and the arthroscopic second look evaluation.</p><p><b>RESULTS</b>Sixty-one (61/70, 87%) cases were available for an average of 13.1 months follow-up assessment postoperatively. There were no significant differences between the RP and control group in functional outcome as evaluated with Lysholm score (96.4 vs. 94.9, P = 0.71) and IKDC grading (cases with A and B gradings: 30 vs. 29, P = 0.586). Regarding objective stability, there were no differences between the 2 group in mean side-to-side difference of KT-1000 (1.69 mm vs. 1.65 mm, P = 0.83), Lachman test (negative cases: 29 vs. 28, P = 1.00) and pivot shift test (negative cases: 31 vs. 27, P = 0.225). There was also no difference between the groups in proprioception evaluation measured with angle repetitive test (4.56° vs. 4.28°, P = 0.522). During second look arthroscopic examination, the grafts synovial coverage rates were found to be 85% in the RP group and 84.2% in the control group, without significant difference (P > 0.05).</p><p><b>CONCLUSIONS</b>Arthroscopic ACL reconstruction with the remnant preserving technique using tendon allograft do not improve the postoperative knee-joint function scores, stability, proprioception and synovial coverage of grafts.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the mechanical changes of the degenerated lumbar disc with finite element analysis.</p><p><b>METHODS</b>A three dimensional finite element model of a human lumbar spine at the L3-L4 disc was established by the software MIMICS and ABAQUS based on computer tomography images. Degeneration was modeled by changes in geometry and material properties. The model was loaded with 0.3 MPa in axial plane. The von Mises stress on the annulus fiber, nucleus pulposus, endplate and facet joints in healthy and degenerated discs was compared.</p><p><b>RESULT</b>Compared with healthy discs, the von Mises stress of disc distributed in the side of annulus fiber, the stress of nucleus pulposus decreased remarkably, the stress of endplate distributed in the posterior part, and the stress of facet joints increased for the degenerated disc.</p><p><b>CONCLUSION</b>The finite element models can provide a method of understanding the relationship between biomechanical performance of the disc due to disc degeneration.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To report the therapeutic effect of reverse fasciocutaneous flap pedicled with perforator branch of anterior medial malleolus artery for soft tissue defect on the dorsal side of foot.</p><p><b>METHODS</b>The perforator branch was located under the guidance of Doppler flowmeter. The flap was designed along the saphenous neurovascular axis. Then the flap was transferred reversely with the perforator branch as rotation point.</p><p><b>RESULTS</b>From Feb. 2002 to Mar. 2008, 12 cases were treated and followed up for 6 - 18 months. All the flaps survived completely. The flap size ranged from 13.5 cm x 3.0 cm to 8 cm x 3 cm. The perforator branch located at 0.5 - 1.5 cm anterior-inferior to the medial malleolus. Both the cosmetic and functional results were satisfactory.</p><p><b>CONCLUSIONS</b>The flap has a reliable blood supply and a flexible design. It is easily performed for soft tissue defect on the dorsal side of foot. It is a new type flap which combined neurocutaneous vascular flap with the perforator flap.</p>

ABSTRACT

<p><b>OBJECTIVES</b>To analyze the risk factors of skin necrosis around incision after total knee arthroplasty, and explore measures for prevention and treatment.</p><p><b>METHODS</b>From January 2003 to November 2005, skin necrosis around incision occurred to 15 out of the 382 cases (494 knees) treated by total knee arthroplasty. The data of the 15 cases were analyzed. Preoperative hospital for special surgery knee score (HSS) was 60 points on average.</p><p><b>RESULTS</b>All wound were healed after debridement. However, the recover time was over 3 weeks longer than the healing time by first intention. Extensor weakness with anterior knee pain appeared among 9 patients, and 6 months later, the symptoms improved obviously. At two years follow-up, the average range of motion was 92 degrees, and the average HSS improved to 86. There were no secondary infection and deformity in the group.</p><p><b>CONCLUSIONS</b>It is the key to prevent skin necrosis around incision during total knee arthroplasty that surgeons must accurately grasp technique of minimally invasive surgery for total knee arthroplasty. To acquire satisfactory outcome, it is imperative to perform debridement as soon as possible while the skin necrosis happens. At the same time, it is important to persist in doing adequate functional exercises throughout the period of treatment.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To assay the expression of KiSS-1 and GnRH in the male rat hypothalamus at different developmental stages, and to explore the significance of KiSS-1 in sex development onset and normal reproduction regulation.</p><p><b>METHODS</b>Expression analyses of KiSS-1 and GnRH genes were conducted in the rat hypothalamus at different developmental stages with RT-PCR and real time-PCR. The testosterone level was assayed by chemoluminescence technique.</p><p><b>RESULTS</b>KiSS-1 mRNA rose gradually during sex development in the rat hypothalamus, highest at puberty and lowered a little at adulthood. KiSS-1 mRNA of the prepubertal, early pubertal, pubertal and adult rats was 1.7, 2.1, 3.5 and 2.0 times higher than that of the infantile rats respectively. The expression of GnRH and KiSS-1 correlated positively (r = 0.905, P < 0.05). But the activation of GnRH neuron was later than KiSS-1. The expression of GnRH was the highest in the puberty rats. GnRH mRNA of the prepubertal, early pubertal, pubertal and adult rats was 1.1, 1.94, 2.42 and 1.92 times higher than that of the infantile rats respectively. The level of testosterone in the adult rats was significantly higher than that at the earlier stage and was the highest at the adult stage.</p><p><b>CONCLUSION</b>The expression of KiSS-1 correlates positively with that of GnRH. KiSS-1 may participate in the regulation of GnRH and is relevant to puberty onset and the regulation of reproduction function.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To introduce the process and outline of fluoroscopy-based navigation system assisted anterior cruciate ligament (ACL) reconstruction, and evaluate its feasibility and accuracy.</p><p><b>METHODS</b>From September 2005 to February 2006, there were 30 cases ACL rupture patients who received fluoroscopy-based navigation system assisted arthroscopy operations for ACL reconstruction (navigation group). At the same time, there were 40 patients who underwent traditional ACL operation (traditional group). For the navigation group, the proper placement of femoral and tibial tunnels was planned preoperatively in standard AP and lateral X-ray view. Intraoperative fluoroscopic images were taken and input into navigation computer system to form the virtual interactive working fields. After placement and registration, signals from patient trackers, which fixed on the distal femur and tibia respectively, and tool trackers, which attached with ACL tibial and femoral guide, were captured by the optic navigation camera and the navigation computer system could pursue the real-time position of the ACL tools and projected into working field to help precise placement of femoral and tibial tunnels. Then results of two groups were observed and evaluated.</p><p><b>RESULTS</b>For navigation group, the mean time extension was 20 min. The tibial tunnel position was measured in all these cases. The tibial tunnel position of navigation group was 45.90% (SD 2.36%), and the traditional group was 41.05% (SD 6.01%). The difference was statistically significant (P < 0.05).</p><p><b>CONCLUSION</b>Fluoroscopy-based navigation system assisted ACL reconstruction improves the accuracy and reproducibility of the tunnel placement.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the rate of systemic inflammatory response syndrome (SIRS) among patients treated by intramedullary nail (IMN) for femoral shaft fracture right after the operation, the effect of femoral nailing on SIRS and sought to differentiate any differences between reamed and unreamed IMN.</p><p><b>METHODS</b>Three hundred and twenty-four patients presenting with acute femoral fractured from April 1997 to April 2005, divided into 2 group depending on ISS. Group 1 had 179 patients (ISS < 16, AIS < 2) including 64 reamed cases and 115 unreamed cases; group 2 had 145 patients (ISS >or= 16, AIS >or= 2) including 51 reamed patients and 94 unreamed. All patients were treated by close reduction and intramedullary nail for fixation. The heart rate, breath rate, temperatures were recorded every 6 h before and after operation. If any of the life sign was above normal, then venous blood samples were taken at once to make sure whether SIRS occurred or not. All data were crosstabs analysis by SPSS 10.0.</p><p><b>RESULTS</b>One hundred and twenty-four patients suffered from SIRS postoperation. In group 1, 24 reamed and 37 unreamed, in group 2, 27 reamed and 36 unreamed. Among those SIRS, 119 cases healed evenly and 5 patients developed further complication. Between reamed and unreamed patients both in group 1 and group 2, there was no significant difference found. The Chi-Square value were 0.159 and 2.885 respectively, total value was 3.467, which P was 0.325, 0.089 and 0.471, all greater than 0.05.</p><p><b>CONCLUSIONS</b>Although SIRS will result from IMN, there is no significant different effect on the rate of SIRS found between reamed and unreamed nail in both group.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To detect the short-term clinical and side effects of cryosurgical ablation for prostate cancer.</p><p><b>METHODS</b>The ENDOCAm cryosurgical system with 2 mm Cryo-probes was used to treat the prostate cancer guided by rectal ultrasonic. 1. Completeness of freeze; 2. Complications; 3. The changes of PSA and PSMA before and after operation were observed respectively.</p><p><b>RESULTS</b>1. The completeness of freeze rate and non-completeness of freeze rate were 34.8% (8/23) and 65.2% (15/23) showed by MRI 3 weeks later after operation; the biopsy results showed that tumor positive rates in completeness of freeze group and non-completeness of freeze group were 0 and 20% (3/15) respectively after 6 months. 2. The incontinence rate was 13%, and all recovered during 1 month; and the erectile dysfunction rate post-operation was 56.2% (9/16). 3. The levels of PSA before and after cryosurgical ablation were (32.98 +/- 35. 50) microg/L, (11.65 +/- 26. 51) microg/L respectively (P < 0.05). PSMA increased significantly after the cryosurgical ablation.</p><p><b>CONCLUSION</b>The short-term curative effects of cryosurgical ablation, which guided by rectal ultrasound for prostate cancer is satisfied with low complication rate and minor vulnus.</p>

ABSTRACT

Ginseng is a valuable medicinal plant with ginsenosides as its mian effective components. Because ginseng is a perennial plant and has a very strict demand for soil conditions, the way of cultivating ginseng by cutting woods is still used in China at present and thus forest resources has been extremely destroyed. Increasing attention has been paid to the hairy roots induced by the infection of Agrobacterium rhizogenes in the production of plant secondary metabolic products for the hairy roots are characterized by rapid growth and stable hereditary and biochemical traits. That has opened a new way for the industrial production of ginseosides. However, there is little report for such studies from China. In this paper, hairy roots of ginseng were induced from the root explants of two-year-old ginseng by Agrobacterium rhizogenes A4 with directly inoculating. The transformed hairy roots could grow rapidly on MS medium and 1/2 MS medium without hormones. The cultured clones of the hairy roots were established on a solid 1/2 MS medium. After 4 - 5 subcultures the hairy roots still maintained a vigorous growth. A pair of primers were designed and synthesized according to the analytical results of RiA4TL-DNA sequence by Slightom et al . 0.8kb rolC was obtained by PCR using the genome DNA of hairy root of ginseng. Transformation was confirmed by PCR amplification of rolC genes from the hairy roots of P. ginseng. Growth rate of hairy roots on liquid medium increased by 2 times then that of the solid medium. The growth of the hairy roots can be divided into three stages: high speed in the first two weeks, middle speed in the 3 - 4 weeks and low speed hereafter. Changing the culture solution at 2 weeks regular intervals is conductive to maintaining the rapid growth of the hairy roots. By means of determination for specific growth rate and ginsenosides content, the high-yield hairy root clone R9923 was selected. The content of monomer gisenoside of Rg1, Re, Rf, Rbl, Rc, Rb2 and Rd in hairy root clone R9923 was determined by the HPLC. The total ginsenosides content in the hairy toot clone R9923 came up to 15.2 mg/g. The suitable culture conditions for ginseng hairy roots growing were 1/2 MS liquid medium (30 g/L glucose), in a shaker at 110 r/min, changing the culture solution at 2 weeks and subculture time 4 weeks. In the liquid fermented culture of 2L medium, the yield of the hairy roots could amount to 270.10 g in 4 weeks. The industrial production of ginsenosides has been preliminarily realized. Effect factors on biomass and ginsenosides content such as culture volume, inoculation, in steps cultural technology at the scale-up process of hairy roots culture were also explorated. Our results have laid a foundation for defining optimum culture manner for large-scale cultivation and large-scale production of ginsenosides.

ABSTRACT

<p><b>AIM</b>To study the chemical structure of SC3, an acidic polysaccharide from Salvia chinesis.</p><p><b>METHODS</b>Based on chemical (including sugar composition analysis, methylation analysis, uronic acid reduction and partial acid hydrolysis) and spectral analysis (IR, NMR, ESI-MS), the structural characterization of SC3 was investigated.</p><p><b>RESULTS</b>SC3 was composed Rha, Ara, Gal and GalA, with its mean molecular weight of 7.7 x 10(4). By means of methylation analysis, partial acid hydrolysis, NMR and ESI-MS spectrum, the linkages and sequence information of SC3 were obtained.</p><p><b>CONCLUSION</b>SC3 is an complicated acidic polysaccharide, obtained for the first time from the plant.</p>
